Tango Therapeutics Achieves 92% Response Rate in Pancreatic Cancer

Tango Therapeutics said its experimental combination of vopimetostat and daraxonrasib produced a 92% objective response rate in pancreatic cancer in early phase 1/2 data, with 11 responses among 12 response-evaluable patients and 9 of those responses confirmed.

The company also reported that 90% of those pancreatic cancer patients were progression free at 6 months, with median progression-free survival not yet reached. Disease control was 100%.

In non-small cell lung cancer, the same combination produced a 100% objective response rate, with all 3 responses confirmed among 3 response-evaluable patients.

The data came from 20 pancreatic cancer patients and 5 lung cancer patients treated in the daraxonrasib arm, out of 59 total patients in the study. More than half of all patients were receiving the combinations as third-line treatment, and the pancreatic cancer group included a heavily pretreated population with 70% carrying liver metastases.

Tango also reported results for its vopimetostat plus zoldonrasib arm in pancreatic cancer. Among 27 response-evaluable patients, the objective response rate was 52%, or 14 of 27 patients, with 10 confirmed responses. Six-month progression-free survival was 74%, and disease control was 96%.

Safety data showed no related grade 4 or 5 adverse events in either combination. In the daraxonrasib arm, the most common treatment-related side effects were rash, stomatitis/mucositis and diarrhea. In the zoldonrasib arm, nausea and vomiting were the most common treatment-related adverse events.

Dose-limiting toxicities were reported only in the daraxonrasib arm at the higher dose level: three DLTs in two patients at vopimetostat 250 mg plus daraxonrasib 100 mg, including one grade 3 rash and one grade 3 stomatitis and fatigue. There were no DLTs at the lower dose level. In the zoldonrasib arm, the company reported no DLTs. There were no discontinuations due to adverse events in either arm.

Tango said it plans to move the vopimetostat plus daraxonrasib combination into phase 3 development in first-line, MTAP-deleted pancreatic cancer. The company also said it intends to initiate a phase 3 randomized-controlled trial in front-line pancreatic cancer and evaluate a second-line path toward registration.
