Disc Medicine's Phase 2 Trial Shows 85% Reduction in Phlebotomy Use

Disc Medicine said its phase 2 Restore-PV trial of disc-3405 cut phlebotomy use sharply in polycythemia vera patients while keeping hematocrit controlled through 26 weeks.

In cohort A, among the 13 patients who completed 26 weeks, mean total phlebotomy events fell to 0.6 over 26 weeks from 4.0 in the 26 weeks before treatment, a drop of 85%. The company said 61.5% of participants remained completely phlebotomy-free after starting treatment, and among the 9 patients who reached the first maintenance period, 77.8% stayed phlebotomy-free during weeks 12 to 32.

The study enrolled 40 adults with PV, split evenly between two cohorts of 20. At the data cut, all 20 patients in cohort A had been dosed and 13 had reached 26 weeks; 18 of 20 patients in cohort B had been dosed.

Disc said disc-3405 increased hepcidin, lowered serum iron and raised ferritin in cohort A. Mean hematocrit stayed below 45% through week 26, and the company said patients showed an initial improvement in symptom burden.

The drug was given subcutaneously at 300 mg every two weeks in cohort A and every four weeks in cohort B after a 12-week dose-escalation period. Disc said the treatment was generally well tolerated, with mild and self-limited injection-site reactions.

The company also pointed to earlier progress in myelofibrosis, saying it presented an encore of phase 2 Rally-MF results for selcodebart and a literature review on anemia in myelofibrosis.
